Benjamin, keep in mind that you will most likely scale them down to something a lot smaller 64x64 or even smaller... The drop shadow will then turn into something like a dark line. Either one of the those is fine as long as you keep the rest of the art consistent - more 3D looking with the inner dropshadow and more cartoony flat without it... Good luck...
i'm all about the drop shadow. but make sure you like how it looks when small, and that diff. images shown in different sizes have appropriate adjustments to the shadow. can also be a super easy way to add depth to your scene, with larger shadows for images farther away from the background... first image looks like a sticker to me.

I like the first one better, and you can easily do it with vectors.
I don't, however, like the drop shadow.
